MAP kinase phosphatase MKP-1 regulates p-ERK1/2 signaling pathway with fluoride treatment

Dental fluorosis is characterized by hypomineralization of tooth enamel caused ingestion excessive fluoride during formation. Excess could have effects on the ERK signaling, which essential for ameloblasts differentiation and development. MAP kinase phosphatase-1 (MKP-1) plays a critical role in regulating related kinases. However, MKP-1 ameloblast mechanisms MKP-1/ERK signaling pathogenesis dental are incompletely understood. Here, we adopted an vitro cell model using murine ameloblasts-like LS8 cells employing sodium (NaF) as inducer. Using this system, demonstrated that exposure led to inhibition p-MEK p-ERK1/2 with subsequent increase expression dose-dependent manner. We further identified, under high dose fluoride, acted negative regulator fluoride-induced leading downregulation CREB, c-myc, Elk-1. Our results identify novel mechanism regulates provide framework studying molecular intervention remodeling normal pathological conditions. inhibitors may prove be benefit therapeutic strategy treatment.
